Jan Schnitzer at Sidney Kimmel Cancer Center is developing vascular targeting antibodies that go to specific organs and tumors. One of their agent is targeted to lung caveolae, which is a little sac in the vascular wall that mediates transfer of material to the lung tissue. Ex vivo studies he previous performed confirmed that about 90% of this agents can be taken up in the lung tissue in the time scale of 30 min. We performed dynamic imaging with the timing interval of 0.5 second to find the finer time scale dynamics of this agent. A rat is positioned diagonally on our gamma camera, while it is injected with 125I tagged this agent. You can see the agent going through the vena cava to the right ventrical of the heart (Image 1), then distributing throughout the body (Image 2), and being taken up mostly in the lungs with a small uptake in liver (Image 3). Jan did not know how quickly this agent got taken up in the lungs, and this imaging provided the much finer time scales that was not obtainable with traditional ex vivo experiments. Also, it was noticed that the liver uptake was higher than what was previously studied. We hypothesized that the uptake of this agent depends on the lung function. So, we compared the images of two rats; one injected under anesthesia, the other injected without anesthesia, which has higher lung function, and this hypothesis was confirmed by comparing this normal uptake in liver of these two animals. This is another advantage of imaging, where the hypothesis can be tested and the result can be obtained quickly and easily.
